Background HemaSpot HF device, Perkin Elmer #226 and Whatman #903 filter paper are commercially available and are designed for collection and storage of capillary blood spotted on filter paper. Laboratory protocol for measles and rubella IgM EIA has been standardized on DBS spotted on Perkin Elmer #226 and Whatman #903filter paper. In view of the fact that different types of filter papers have different adsorption qualities, the mesh of filter paper might behave differently during elution of sample from it, no standardize elution protocol available for measles rubella IgM testing for HemaSpot HF device, it’s important to first standardize the elution protocol of DBS collected on HemaSpot HF device and its use for measles and rubella IgM assay using Euroimmun kit before it could be field tested in surveillance program.
Objectives Standardize the elution of serum from dried blood sample (DBS) collected on HemaSpot HF devices for measles and rubella IgM enzyme immune assay (EIA) taking IgM titers in serum sample as gold standard.
Study Methods The proposed study is a laboratory-based study that will involve laboratory test experiments using various dilution factors of sample diluent to standardize the elution protocol for measles and rubella IgM EIA from DBS spotted on HemaSpot HF device, Perkin Elmer #226 and Whatman #903filter paper. g. The study requires maximum four IgM positives for measles and four for rubella. Two in the range of moderate to high titers and two in borderline to moderate. High titer sera can be diluted with negative sera to obtain the desired borderline range. To test the various dilutions of buffer for elution of DBS, tests will be conducted in duplicate in three different test experiments. a. The results of DBS eluted from HemaSpot HF devices, Perkin Elmer #226 and Whatman #903filter paper in different dilutions, will be compared with its corresponding serum sample.
Expected outcomes and use of results Outcome of the study is to have a standardized protocol for elution od serul from DBS spotted on different types of filter paper cards commercially available for conducting measles and rubella diagnostic EIAs. Based on the results of this study, a field study will be designed to demonstrate usefulness of DBS collection for laboratory confirmation of measles and rubella suspected cases in surveillance program.
